FSTHED (Fluorescence Sensitivity Tunable Holographic Excitation Device) is a novel optical device used for ultra-sensitive imaging and spectral imaging with unprecedented fluorescence sensitivity. It consists of two main components: an optical desk and a nanostructured holographic element. The optical desk illuminates the sample and directs the incident light beam to the holographic element. The holographic element comprises of a nanostructured aluminium oxide membrane, i.e. aluminium oxide (Al2O3), patterned with a specially designed pattern to maximize light-tissue interaction. The combination of the light-tissue interaction and the specially designed pattern results in a highly sensitive fluorescence image.
FSTHED is designed to be used in the ultra-sensitive bio-imaging applications that require fluorescence imaging with precision and accuracy. The imaging applications include, but are not limited to, single-cell imaging, tissue fluorescence imaging, disease diagnosis, medical imaging, and drug delivery. FSTHED is also applicable for spectral imaging, which can be used to acquire spectra of chemo-signaling components or species in living cells or tissues. Spectral imaging techniques, such as Stimulated Emission Depletion (STEM) or Resonance Energy Transfer (RET), provide a powerful tool to detect subtle changes in the bio-chemistries of live cells or tissues over time.
The working principle of FSTHED is based on the underlying principles of holography. Holography is a technique used to store, transmit, and reconstruct wavefronts of light. It can also be used to encode, manipulate, and store optical information. The working of FSTHED is based on the holographic reconstruction of two-dimensional optical field patterns generated using a combination of the nanostructured aluminium oxide membrane and the incident light beam.
